The South Zone of the Ranji Trophy, 2000-01 saw the Tamil Nadu, Hyderabad and Karnataka teams becoming the toppers.

6 teams participated under the South Zone of the Ranji Trophy, 2000-01, all the regular ones mainly coming from the Southern region of India. The teams were representing different states from the Southern region of India in the most popular and top class domestic cricket tournament in India. The participating teams included the likes of Tamil Nadu, Goa, Andhra, Kerala, Karnataka and Hyderabad and they played against each other for claiming the top spot in the zone. Securing a berth in the knock out level of the Ranji Trophy, 2000-01 was also another principal objective of all the teams. Only the teams like Tamil Nadu, Hyderabad and Karnataka could fulfill this objective from the South Zone, as they earned the highest points. The Tamil Nadu, Hyderabad and Karnataka teams earned total of 26, 25 and 24 points, respectively from the zonal level league matches.

The match between the teams of Andhra and Kerala was the first of the South Zone league matches in the Ranji Trophy, 2000-01 and the match was played at Vijayawada from 01-04 November, 2000. The match remained drawn during the specified period of time and the match officials awarded the Kerala and Andhra teams 5 and 3 points, respectively for the match. The Karnataka team played against the Hyderabad team in the second match of the South Zone and the match was held at Bangalore from 01-04 November, 2000. The drawn match saw the Karnataka team earning 5 points and Hyderabad earning 3 points from it. The third match of the zone involved the teams like Goa and Tamil Nadu and the two teams played the match at Goa from 01-04 November, 2000. The winner of the match was the Tamil Nadu team with a margin of 203 runs and the team earned 8 points from the match. The Goa team failed to earn any points from the match

The Tamil Nadu team appeared in the fourth of the South Zone league matches in the Ranji Trophy, 2000-01 also and this time, it played against the Karnataka team at Chennai from 08-11 November, 2000. The Tamil Nadu and Karnataka teams earned 5 and 3 points, respectively from the match, as it remained drawn during the specified period of time. The next match of the zone involved the teams like Hyderabad and Kerala and the match took place at Hyderabad from 08-11 November, 2000. The Hyderabad team won the match by a margin of an innings and 46 runs for earning 8 points and the Kerala team failed to earn any points from the match. The Goa and Andhra team met with each other in the next match of the zone and the match was held at Margao from 09-12 November, 2000. The match was won by the Andhra team with a margin of an innings and 20 runs to earn 8 points and the Goa team remained pointless in the match.

The Karnataka team played against the Andhra team in the seventh of the South Zone league matches in the Ranji Trophy, 2000-01 and the match took place at Belgaum from 15-18 November, 2000. The Karnataka team earned 8 points from the match by clinching a victory of 5 wickets and the Andhra team failed to earn any points from the match. The next match of the South Zone saw the teams of Hyderabad and Tamil Nadu playing against each other at Hyderabad from 15-18 November, 2000. The match remained drawn during the stipulated duration and the Tamil Nadu and Hyderabad teams earned 5 and 3 points, respectively from the match. The teams of Kerala and Goa then met with each other in the next match of the zone, which was held at Cochin from 15-18 November, 2000. The Kerala and Goa teams earned 5 and 3 points, respectively from the drawn match.

The tenth of the South Zone league matches in the Ranji Trophy, 2000-01 involved the teams like Hyderabad and Goa and the two teams played the match at Hyderabad from 22-25 November, 2000. The Hyderabad team played very well in the match and clinched a victory of 153 runs for earning 8 points from it. The Goa team remained pointless in the match. The next match of the zone saw the teams of Tamil Nadu and Andhra playing against each other at Chennai from 22-25 November, 2000. The match remained drawn during the specified period of time and the match officials awarded both the Tamil Nadu and Andhra teams 3 points, each for the match. The Kerala team then played against the Karnataka team in the next match of the zone and the match took place at Cochin from 22-25 November, 2000. The Kerala team earned 8 points from the match by clinching a huge victory of 10 wickets and the Karnataka team failed to earn any points from the match.

The Goa and Karnataka teams met with each other in the next match of the South Zone and the two teams played the match at Margao from 29 November-02 December, 2000. The Karnataka team played very well in the match and earned 8 points from it by clinching a victory of 9 wickets. The Goa team could not earn any points from the match. The next match of the zone involved the teams like Andhra and Hyderabad and the match was held at Kurnool from 29 November-02 December, 2000. The Andhra team won the drawn match on the basis of its first innings lead and earned 5 points from it. The Hyderabad team also earned 3 points from the match. The last of the South Zone league matches in the Ranji Trophy, 2000-01 was played between the teams of Kerala and Tamil Nadu and the two teams played the match at Cochin from 29 November-02 December, 2000. The match remained drawn during the stipulated duration and the Tamil Nadu and Kerala teams earned 5 and 3 points, respectively from the match.
